Being developed to produce large reductions in body weight in adults with obesity, or overweight with weight-related complications, and to improve glycaemic control in type 2 diabetes. Secondary development claims cover knee osteoarthritis pain and obstructive sleep apnoea in people with obesity. None of these claims has been reviewed or accepted by any regulator.
It is not approved anywhere in the world. Eli Lilly had not filed for approval as of October 2026 and stated it planned to submit in the first quarter of 2027; no FDA, EMA or MHRA filing has been accepted and no PDUFA date exists. Lilly’s own release states retatrutide is "legally available only to participants in Lilly’s clinical trials."
The weight reductions reported in the phase 3 TRIUMPH programme are the largest in this cluster, and every one of those trials was designed, funded, monitored and analysed by Lilly, with Lilly employees in the author lists. TRIUMPH-3 enrolled people with established cardiovascular disease and recorded in-study cardiovascular event counts; it was not powered as a cardiovascular outcomes trial and did not show a cardiovascular benefit.
Retatrutide is sold online by "research peptide" vendors. Those products are not the studied product: they have no verified identity, purity, potency, sterility or endotoxin testing, no regulatory oversight and no chain of custody. The trial data below describe a specific sponsor-manufactured molecule given under medical supervision, and say nothing about what is in a vial bought online.
At a glance
A single lipidated peptide that agonises three receptors at once: GLP-1, GIP and glucagon. GLP-1 and GIP agonism act on appetite regulation and insulin secretion; glucagon-receptor agonism is intended to add an energy-expenditure and hepatic-fat component on top of appetite suppression. Albumin binding via the lipid side chain extends the half-life enough for once-weekly administration. Human trials confirm the expected downstream results — weight loss, HbA1c reduction, liver-fat reduction — and GLP-1 and GIP effects are well established in humans from the marketed drugs of that class. What has not been done is a human dose-dissection study isolating the glucagon-receptor arm, so the specific contribution of the third receptor, which is the whole rationale for the molecule, remains inferred rather than demonstrated.
Dated determinations, each flagged for whether it is final and whether it covers safety only. A safety review is not an efficacy finding, and most reporting in this category conflates the two.
As of 13 October 2026 Eli Lilly had not filed. In its 23 July 2026 release the company stated it "plans to subsequently submit retatrutide in Q1 2027 for U.S. approval," as a Biologics License Application, attributing the delay to completing the Chemistry, Manufacturing and Controls data package. No FDA, EMA or MHRA filing has been accepted and no PDUFA date exists. Lilly’s own TRIUMPH-1 release states retatrutide is "legally available only to participants in Lilly’s clinical trials." isFinal is false because nothing has been decided — this is the absence of a determination, recorded so that the absence is visible.
